Unlocking dragonmarks is done on a per-server basis by completing the Hall of the Mark quest by Lockania in The Marketplace near the auctioneer.
You can start your character with a least dragonmark, upgrade to lesser at level 6 and greater at level 9.

Lesser: 2 +1 Least
Greater: 1 +1 Least +1 Lesser
You can spend Enhancement Points and get upward of 4 additional uses of each dragonmark per day.
+1 per day at level 2, min 3 points spent.
+2 per day at level 6, min 18 points spent.
+3 per day at level 10, min 33 points spent.
+4 per day at level 14, min 48 points spent.
If you went all out and loaded up on dragonmarks and enhancements, you could have the following daily dragonmark loadout:
Least: 9 | Lesser: 7 | Greater: 5
You may only posess 1 dragonmark line at any time.
Warforge and Drow may not gain dragonmarks.
Loading a halfling of any class with 3 feats and 10 enhancement points could conceivably solo for most of their career or offset 495 spell points for a healer.
Taking the two spell points feats for casters (Mental Toughness and Improved Mental Toughness) would only grant a total of 210 spell points at level 20, so for an additional feat and 10 enhancement points, you’re more than double the spell points AND still your enhancement lines for bonus class-based and race-based spell points.
